Generally yes, you do need permission from both store managers (your current one, as well as the one at the store that you want to transfer to) in order to get a transfer to go through. However, if you truly are going to be forced to quit due to your current store manager not allowing the transfer, then I would suggest going to the store that you want to transfer to and explain your situation to the store manager/co-manager over HR at that location and if he/she believes you'd be an asset at that store, then he/she may make a phone call or two in order to make it happen. Not all store management is bad/uncaring... some actually will try to help if the situation calls for it.

This happened to the guy I replaced (night manager). Basically he wanted to leave our store because he hated the (then new) Store Manager, our previous store manager had went to a different store and he wanted the Night Manager to go with him. Our then Store Manager held it up out of spite until our previous Store Manager called someone at District and the move got forced through. Our store manager at the time was livid because he took everything personal and saw it as a slap in the face.
Just contact the District Hr Coordinator with valid reasons they can not dispute (financial, work-life, personal, etc). Store Managers at times think they are GODS...
